Reactions of Flavonoids with o‑Quinones Interfere with the Spectrophotometric Assay of Tyrosinase Activity

Flavonoids are important food components with antioxidant properties and many of them have been described as tyrosinase inhibitors. Oxidation of quercetin, kaempferol, morin, catechin, and naringenin by mushroom tyrosinase and their influence on the oxidation of l-dopa and l-tyrosine was studied. Reaction rates measured spectrophotometrically and by oxygen consumption differed substantially. All tested flavonoids reacted with 4-tert-butyl-o-benzoquinone and/or 4-methyl-o-benzoquinone, although at different rates. Mechanisms of interference of p-diphenols with the Trinder reaction

p-Diphenols, such as homogentisic acid, gentisic acid, etamsylate, and calcium dobesilate, interfere with diagnostic tests utilizing the Trinder reaction but the mechanisms of these effects are not fully understood. We observed substantial differences both in oxidation of p-diphenols by horseradish peroxidase and their influence on oxidation of 4-aminoantipyrine and various phenolic substrates. Homogentisic acid was rapidly oxidized by the enzyme and completely blocked chromophore formation. Enzymatic oxidation of the remaining p-diphenols was slow and they only moderately inhibited chromophore formation. This data was collected during an short-term incubation experiment in March 2017 that investigated the response of a surface plankton community to upwelling. This experiment was carried in the framework of the SFB754-funded KOSMOS mesocosm study that took place in La Punta, Callao, Peru between February-April 2017. A total of six different treatments were used to disentangle chemical and biological characteristics of deep water that influence surface plankton blooms: 2 different deep water sources with different nutrient concentrations; 3 treatments to distinguish the effects of inorganic nutrients, organic nutrients and deep water microbial populations.
